CCX733 is a small molecule antagonist of the **C-X-C chemokine receptor type 7 (CXCR7)**, also known as **atypical chemokine receptor 3 (ACKR3)**. Developed by **ChemoCentryx**, it targets the CXCR4-CXCR7-CXCL12 axis, which is implicated in the progression, metastasis, and survival of various cancers, including renal cell carcinoma (RCC). CXCR7 is a high-affinity receptor for the chemokines CXCL12 (SDF-1) and CXCL11 (I-TAC). Unlike the related receptor CXCR4, CXCR7 does not typically signal through classical G-protein pathways but acts as a scavenger for its ligands or as a modulator of CXCR4 signaling. Preclinical studies have shown that CCX733 can influence cell migration and AKT phosphorylation in human renal cancer cell lines, although its functional effects can vary between inhibition and enhancement of migration depending on the specific cellular context and the relative expression of CXCR4 and CXCR7.
