Overview

CCX872-B is an orally administered, potent and selective small molecule antagonist of the chemokine receptor CCR2 (C-C chemokine receptor type 2). By blocking CCR2, it inhibits the binding of its ligand CCL2 (monocyte chemoattractant protein-1 or MCP1), thereby preventing CCR2 activation and downstream signal transduction. This mechanism results in reduced recruitment of monocytes/macrophages to sites of inflammation or tumor microenvironments, leading to immunomodulatory effects such as decreased tumor-associated macrophages and regulatory T cells, increased cytotoxic T cell infiltration, inhibition of inflammatory processes, angiogenesis, tumor cell migration and proliferation. CCX872-B has demonstrated antineoplastic activity in preclinical models and has been evaluated clinically for pancreatic cancer in combination with FOLFIRINOX chemotherapy. The drug was developed by ChemoCentryx.
