CCX9588 is a selective, orally bioavailable small molecule antagonist of the C-C chemokine receptor type 1 (CCR1). Developed by ChemoCentryx, CCX9588 is designed to block the binding of CCR1 ligands, such as CCL3 (MIP-1α) and CCL5 (RANTES), thereby inhibiting the trafficking and recruitment of myeloid-derived suppressor cells (MDSCs) to the tumor microenvironment. In preclinical models of triple-negative breast cancer, CCX9588 has demonstrated synergistic anti-tumor and anti-metastatic effects when combined with immune checkpoint inhibitors, such as anti-PD-L1 antibodies, by reducing immunosuppressive MDSCs and increasing CD8+ T-cell infiltration. It has also shown potential in multiple myeloma by inhibiting the dissemination of plasma cells in the bone marrow microenvironment.
