The CD103+cDC1 vaccine (cDCV) is an experimental, cell-based cancer immunotherapy consisting of in vitro-generated type 1 conventional dendritic cells (cDC1s) that express the CD103 surface marker. The vaccine is prepared by loading these dendritic cells with tumor cell lysates (such as those derived from osteosarcoma) and activating them with the TLR3 agonist poly-I:C. Its primary mechanism of action involves the cross-presentation of tumor antigens to CD8+ T cells, which promotes the infiltration of cytotoxic T lymphocytes into the tumor microenvironment. Preclinical research has demonstrated that the vaccine can induce potent anti-tumor immunity in models of osteosarcoma and melanoma, particularly when used in combination with immune checkpoint blockade therapies like anti-CTLA-4. The development of this vaccine is currently in the preclinical/academic research stage, with findings published in peer-reviewed journals by academic investigators.
