A CD103-neutralizing antibody is a monoclonal antibody designed to bind to and inhibit the activity of CD103 (Integrin alpha E), a protein that forms the heterodimeric integrin alphaEbeta7. CD103 is predominantly expressed on intraepithelial lymphocytes and specific subsets of dendritic cells (CD103+ DCs) that are crucial for maintaining mucosal immunity and peripheral tolerance. These dendritic cells facilitate the induction of Foxp3+ regulatory T cells (Tregs) through mechanisms involving retinoic acid production. In preclinical models of systemic lupus erythematosus (SLE), such as the NZBxNZWF1 mouse strain, neutralization of CD103 has been shown to exacerbate disease severity and increase mortality, suggesting that CD103+ cells play a protective role in the regulation of autoimmune responses. This type of antibody is primarily utilized as a research tool to investigate the immunoregulatory functions of CD103-expressing cells in various inflammatory and autoimmune contexts.
