CD117 CAR-iNKT-IL15 cells are an experimental, off-the-shelf allogeneic cell therapy designed for the treatment of acute myeloid leukemia (AML). This therapy utilizes invariant natural killer T (iNKT) cells, a unique population of innate-like T cells that recognize glycolipids presented by CD1d and are known for their ability to mediate antitumor activity without inducing graft-versus-host disease (GVHD). The cells are engineered with a chimeric antigen receptor (CAR) targeting CD117 (c-Kit), a receptor tyrosine kinase highly expressed on AML blasts. To overcome functional exhaustion and enhance persistence, the CAR construct incorporates interleukin-15 (IL-15). Preclinical research conducted at Stanford University indicates that these cells can effectively eliminate CD117-high AML cells and CD1d-positive tumor-associated macrophages, potentially reversing the immunosuppressive tumor microenvironment associated with aggressive leukemia.
