CD123-CD16 bispecific antibody-modified NK cells are donor-derived natural killer (NK) cells that have been modified using a bispecific antibody targeting both CD123 on tumor cells and CD16 on NK cells. The mechanism leverages the bispecific antibody to bind CD123 (interleukin-3 receptor α chain) present on acute myeloid leukemia (AML) cells and CD16 (Fcγ receptor IIIa) on NK cells, bringing them into close proximity to induce targeted cytotoxicity through antibody-dependent cell-mediated cytotoxicity (ADCC). This investigational immunotherapy aims to harness and enhance the innate cytotoxic potential of NK cells against CD123-positive hematological malignancies, primarily relapsed or refractory acute myeloid leukemia (AML). The therapy is currently being tested in early-phase clinical trials and involves ex vivo modification and expansion of NK cells prior to infusion into patients[1][4][6].
