Overview

CD123-CD33 cCAR T cells is an autologous cell therapy developed by iCell Gene Therapeutics using its proprietary compound CAR (cCAR) platform. This therapy involves engineering a patient's own T cells to express two distinct chimeric antigen receptors (CARs) on a single cell, targeting both CD123 (Interleukin-3 receptor subunit alpha) and CD33 (Myeloid cell surface antigen CD33). Both antigens are frequently overexpressed in myeloid malignancies, including acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), and myeloproliferative neoplasms (MPN), and are present on leukemic stem cells. By targeting two antigens simultaneously, the cCAR design aims to enhance therapeutic efficacy and mitigate the risk of disease relapse caused by antigen escape or tumor heterogeneity. The therapy is currently being evaluated in Phase I clinical trials for patients with relapsed or refractory high-risk hematologic malignancies.
