CD123 chimeric antigen receptor memory-like natural killer (CD123 CAR ML NK) cells are an experimental cell therapy being developed for the treatment of acute myeloid leukemia (AML). This approach utilizes "memory-like" NK cells, which are generated by brief ex vivo activation with a cytokine cocktail consisting of IL-12, IL-15, and IL-18. These pre-activated NK cells exhibit enhanced effector functions, including increased cytokine production and cytotoxicity, as well as improved persistence compared to conventional NK cells. To provide antigen-specific targeting, these memory-like NK cells are genetically engineered to express a chimeric antigen receptor (CAR) directed against CD123 (the alpha chain of the interleukin-3 receptor), a protein frequently overexpressed on AML blasts and leukemic stem cells. The therapy aims to combine the potent, long-lasting anti-tumor activity of memory-like NK cells with the precision of CAR-mediated targeting to provide a more durable response in patients with relapsed or refractory AML.
