CD123 SIR-T is an investigational Synthetic Immune Receptor T-cell (SIR-T) therapy being developed by Angeles Therapeutics for the treatment of hematologic malignancies. The therapy utilizes a proprietary Synthetic Immune Receptor (SIR) designed to target CD123 (Interleukin-3 receptor alpha chain), a protein frequently overexpressed on the surface of leukemic blasts and leukemic stem cells, particularly in acute myeloid leukemia (AML) and blastic plasmacytoid dendritic cell neoplasm (BPDCN). Unlike traditional CAR-T cells, which use a single-chain variable fragment (scFv) fused to signaling domains, SIR-T cells incorporate a synthetic receptor that leverages the natural T-cell receptor (TCR) signaling machinery. This design is intended to provide enhanced sensitivity to low-antigen density and improved metabolic fitness, potentially leading to better persistence and efficacy in the clinical setting. The program is currently in the preclinical stage of development.
