CD123 targeted CAR-NK cells are genetically engineered natural killer (NK) cell therapies designed to express a chimeric antigen receptor (CAR) that specifically targets the interleukin-3 receptor alpha chain (CD123), which is overexpressed on leukemic stem cells and myeloid blasts in acute myeloid leukemia (AML) and blastic plasmacytoid dendritic cell neoplasm (BPDCN). The therapy aims to enhance NK-cell antitumor effects while reducing toxicity compared to other cellular therapies. Preclinical studies have demonstrated potent antigen-dependent activation and cytotoxicity against AML both *in vitro* and *in vivo*. Clinical trials are ongoing for relapsed/refractory AML and BPDCN[1][2][4][5].
