CD123.ENG.iC T cells are an experimental cell-based immunotherapy consisting of T cells genetically modified to secrete bispecific T-cell engagers (ENG) targeting CD123 (interleukin-3 receptor alpha) and to express an inducible CD40 (iC) co-stimulatory molecule. The CD123-specific engager molecule typically consists of two single-chain variable fragments (scFvs) that bind CD123 on tumor cells and CD3 on T cells, facilitating an immune synapse. The inducible CD40 component is designed to be activated by a chemical inducer of dimerization (CID), such as rimiducid, to provide a co-stimulatory signal intended to enhance T-cell persistence and anti-tumor activity. Developed primarily for the treatment of acute myeloid leukemia (AML), preclinical studies have compared this construct to other variants, such as those incorporating MyD88 signaling (iMC), to optimize effector function.
