Overview

CD123CAR-DOT is an allogeneic, next-generation cellular immunotherapy composed of Delta One T cells (DOTs) genetically engineered to express a chimeric antigen receptor (CAR) targeting CD123, the interleukin-3 receptor alpha chain, which is highly expressed on acute myeloid leukemia (AML) blasts. These DOTs are a proprietary Vδ1+ γδ T cell product known for robust cytotoxicity and upregulation of multiple activating and cytotoxicity-associated NK cell receptors. Retroviral transduction of DOTs with a CAR specific for CD123 greatly augments their ability to eliminate AML cells both in vitro and in vivo. CD123CAR-DOTs release type 1 antitumor cytokines and chemokines upon recognizing AML cells, demonstrating potent and sustained anti-leukemic effects in preclinical models. Their allogeneic, HLA-independent nature enables an "off-the-shelf" therapeutic approach for AML. IL-15 co-administration further enhances their efficacy and persistence[1][3].
