CD127 antibodies are a class of monoclonal antibodies designed to target and inhibit the alpha chain of the interleukin-7 receptor (IL-7Rα), commonly known as CD127. This receptor is a critical component of the signaling complexes for interleukin-7 (IL-7) and thymic stromal lymphopoietin (TSLP), which are essential for the development, maintenance, and proliferation of T cells and innate lymphoid cells (ILCs). By antagonizing CD127, these therapeutic agents aim to suppress pathogenic immune responses in autoimmune and inflammatory conditions, such as ulcerative colitis, Sjögren's syndrome, and multiple sclerosis, without broadly depleting the immune system. Furthermore, CD127 is frequently overexpressed in T-cell acute lymphoblastic leukemia (T-ALL), making it a target for potential antineoplastic therapies. Several specific assets, including lusvertikimab (OSE-127) and BMS-986227, have entered clinical development to explore these indications.
