CD133 CAR-LH is a second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target CD133-positive glioblastoma (GBM) brain tumor initiating cells (BTICs). Developed by researchers at McMaster University and the University of Toronto, the construct utilizes a single-chain variable fragment (scFv) derived from a humanized CD133-specific monoclonal antibody. The "LH" designation refers to the specific orientation of the scFv, where the light chain is linked to the heavy chain (Light chain-linker-Heavy chain). The CAR includes a CD8a transmembrane domain, a CD28 costimulatory domain, and a CD3ζ signaling tail. Preclinical studies have demonstrated that these CAR-T cells can selectively recognize and eliminate CD133+ GBM cells, which are often associated with treatment resistance and disease recurrence.
