CD133-specific BiTE is a preclinical bispecific T-cell engager (BiTE) designed to target CD133 (Prominin-1), a marker for brain tumor initiating cells (BTICs) in glioblastoma. The molecule consists of two arms: one specific to CD133 (derived from the RW03 monoclonal antibody) and the other specific to the CD3 antigen on T cells. By cross-linking CD133-expressing tumor cells with T cells, the BiTE induces T-cell activation and directed cytotoxicity against the tumor cells. It was developed by researchers at McMaster University and the University of Toronto to address treatment-refractory and recurrent glioblastoma, demonstrating the ability to redirect T cells to kill glioblastoma cells with high efficiency in preclinical models.
