CD133-targeted oncolytic adenovirus (CD133-OAd) is an engineered oncolytic viral therapy designed to selectively target and eliminate CD133-positive cancer stem cells. Developed by researchers at the University of Minnesota, this adenovirus-based agent utilizes a library-screened targeting mechanism to infect cells expressing CD133 (Prominin-1), a marker associated with treatment resistance, recurrence, and metastasis in various cancers, including colorectal cancer (CRC). By lysing these cells, CD133-OAd aims to overcome resistance to standard chemotherapy and radiotherapy. Preclinical studies in mouse models of peritoneal metastatic colon cancer have demonstrated significant suppression of tumor growth and prolonged survival without severe toxicity, particularly when used in combination with standard-of-care treatments.
