Overview

CD133 tumor antigen vaccine refers to a class of cancer immunotherapies, most notably **ICT-121**, designed to target cancer stem cells (CSCs) expressing the CD133 (prominin-1) glycoprotein. ICT-121 is an autologous dendritic cell (DC) vaccine where a patient's own monocytes are harvested via apheresis, matured into dendritic cells, and pulsed with purified peptides derived from the CD133 antigen. These engineered cells are then re-administered to the patient (typically via intradermal injection) to prime the immune system to recognize and eliminate CD133-positive tumor cells. This approach specifically targets the CSC population, which is often resistant to conventional chemotherapy and radiation and is responsible for tumor recurrence and metastasis. ICT-121 was developed by ImmunoCellular Therapeutics and reached Phase I/II clinical trials for recurrent glioblastoma multiforme (GBM) in HLA-A2 positive patients, though development was ultimately terminated.
