Overview

CD137L-DC-EBV-VAX is an investigational autologous dendritic cell (DC) vaccine designed for immunotherapy of Epstein-Barr virus (EBV)-associated cancers, particularly nasopharyngeal carcinoma (NPC). The therapy uses monocyte-derived dendritic cells differentiated via reverse signaling through the CD137 ligand (CD137L), which enhances their potency in stimulating T-cell responses. These DCs are then pulsed with EBV antigens to specifically target and induce cytotoxic T lymphocyte responses against EBV-positive tumor cells. In phase I clinical trials, the vaccine demonstrated a favorable safety profile and induced anti-tumor immune responses, including partial tumor regression and prolonged progression-free survival in some patients with locally recurrent or metastatic NPC[1][3][5][9].
