CD146 CAR-T cells + GPC3 CAR-T cells is a sequential chimeric antigen receptor (CAR) T-cell therapy being developed by Essen Biotech for the treatment of advanced relapsed or refractory ovarian cancer, including ovarian clear cell carcinoma (OCCC). The therapeutic regimen involves lymphodepleting chemotherapy with cyclophosphamide and fludarabine, followed by the sequential intravenous infusion of autologous CAR-T cells targeting CD146 (Arm A) and subsequently CAR-T cells targeting Glypican-3 (GPC3) (Arm B). CD146 (Melanoma Cell Adhesion Molecule) and GPC3 are both highly expressed in various solid tumors, and this dual-targeting sequential approach is designed to address tumor heterogeneity and enhance the anti-tumor response in patients with advanced ovarian malignancies.
