CD146-targeted CAR-T cell therapy is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed by Essen Biotech for the treatment of advanced relapsed or refractory ovarian cancer. The therapy consists of patient-derived T cells that have been genetically modified to express a CAR targeting CD146 (also known as Melanoma Cell Adhesion Molecule, MCAM), a cell surface glycoprotein associated with tumor progression and metastasis in various solid tumors. In clinical development, this therapy is evaluated as part of a sequential treatment regimen following lymphodepleting chemotherapy with cyclophosphamide and fludarabine. The regimen involves the infusion of CD146-directed CAR-T cells followed by the administration of GPC3-directed CAR-T cells, a strategy intended to address tumor heterogeneity and improve therapeutic outcomes in patients with advanced ovarian cancer.
