CD155-BBz CAR-T cells are a type of chimeric antigen receptor T cell therapy engineered to target the CD155 antigen, which is highly and homogeneously expressed in various digestive system cancers such as gastric, esophageal, pancreatic, and colon cancers. The construct uses the extracellular domain of human TIGIT for antigen recognition and incorporates 4-1BB (CD137) and CD3ζ signaling domains (BBz) to enhance T-cell activation and persistence. Preclinical studies have demonstrated that these CAR-T cells mediate significant antitumor activity in vivo, inducing complete tumor regression and long-lasting immunologic memory in solid tumor xenograft models. This approach aims to overcome barriers posed by cancer cell heterogeneity and immunosuppressive microenvironments—particularly those involving M2-like tumor-associated macrophages expressing CD155—making it a promising candidate for solid tumor immunotherapy[1].
