Overview

CD155 CAR-T cells represent an innovative cellular therapeutic strategy in cancer immunotherapy. These are T cells genetically engineered with a chimeric antigen receptor (CAR) designed to specifically recognize and bind to CD155, also known as the Poliovirus Receptor (PVR). CD155 is an adhesion molecule that is frequently overexpressed on the surface of various cancer cells, including those in acute myeloid leukemia (AML) and numerous solid tumors, while being minimally expressed in normal tissues. The binding of the CAR-T cells to CD155 on tumor cells triggers the activation of the CAR-T cells, leading to potent antitumor activity and the destruction of the cancer cells. Furthermore, CD155 plays a role in tumor immune evasion by interacting with inhibitory receptors like TIGIT on immune cells, and targeting CD155 with CAR-T cells can help overcome this immunosuppression, thereby enhancing the body's natural ability to fight cancer. Preclinical studies have demonstrated significant reduction in tumor burden across various hematological and solid tumor models, suggesting a broad therapeutic potential for this therapy.
