CD16-enhanced HSC-engineered NKT cells are an allogeneic, off-the-shelf cellular immunotherapy developed using a hematopoietic stem cell (HSC) engineering platform. These cells are genetically modified to express an invariant natural killer T (NKT) cell receptor (TCR) and a high-affinity, non-cleavable CD16a (FcγRIIIa) receptor. This dual-targeting approach allows the cells to recognize tumors via CD1d-presented glycolipid antigens and to mediate potent antibody-dependent cellular cytotoxicity (ADCC) when used in combination with tumor-specific monoclonal antibodies. Additionally, these cells are designed to deplete immunosuppressive tumor-associated macrophages (TAMs) and myeloid-derived suppressor cells (MDSCs) within the tumor microenvironment. The platform utilizes a scalable, feeder-free in vitro differentiation system to produce high-purity NKT cells with superior metabolic fitness and persistence compared to peripheral blood-derived counterparts.
