CD171 specific CAR T cells expressing EGFRt are a form of chimeric antigen receptor (CAR) T cell therapy engineered to target the neural cell adhesion molecule L1CAM (also known as CD171), which is overexpressed in several solid tumors, including neuroblastoma and certain brain cancers. These autologous or allogeneic T cells are genetically modified to express a synthetic receptor that recognizes the CD171 antigen on tumor cells, enabling targeted cytotoxicity. The construct also includes expression of truncated epidermal growth factor receptor (EGFRt), which serves as a selectable marker and potential safety switch for tracking or depleting the infused CAR-T cells if needed. This dual engineering approach aims to enhance specificity and safety in targeting tumors with high levels of CD171 while allowing for clinical control via the EGFRt component.
