CD180-ADC is an antibody-drug conjugate (ADC) designed to target CD180 (also known as RP105), a type I single-pass transmembrane protein that is highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells. Developed by Corellia AI and Champions Oncology, the agent consists of a fully human monoclonal antibody specific for the CD180/MD-1 complex, conjugated to a topoisomerase I inhibitor payload with a drug-to-antibody ratio (DAR) of 8. Upon binding to CD180, the ADC is rapidly internalized, releasing the cytotoxic payload to inhibit topoisomerase I, leading to DNA damage and apoptosis in malignant cells. Preclinical data presented at AACR 2026 demonstrated potent cytotoxicity across AML models and a favorable safety profile in non-human primates, supporting its advancement into clinical trials.
