Drug intelligence / Profile preview
CD19×BCMA×CD3 trispecific antibody (ALX002) is an AI-optimized, dual-targeted T-cell engager (TCE) developed by Ailux (a subsidiary of XtalPi) for the treatment of B cell-mediated autoimmune diseases, including systemic lupus erythematosus (SLE) and rheumatoid arthritis. The antibody is designed to simultaneously target CD19 on B cells and BCMA on plasma cells, enabling deep and broad depletion of the B-cell lineage to achieve a meaningful 'immune reset'. ALX002 incorporates a proprietary CD3-masked design that limits baseline CD3 binding and T-cell activation in the absence of target B cells, thereby reducing the risk of cytokine release syndrome (CRS) and providing a wider therapeutic window. Preclinical studies have demonstrated potent B-cell depletion and low, transient cytokine release, supporting its clinical development with IND filings planned for 2027.
