Overview

CD19-7x19 CAR-T + tislelizumab is a combination therapy consisting of a fourth-generation autologous chimeric antigen receptor (CAR) T-cell therapy and the anti-PD-1 monoclonal antibody tislelizumab. The CAR-T component is engineered to target the CD19 antigen and co-express Interleukin-7 (IL-7) and Chemokine (C-C motif) ligand 19 (CCL19). This design, often referred to as a 4SCAR (fourth-generation signaling CAR), aims to enhance the infiltration, accumulation, and survival of CAR-T cells within the tumor microenvironment, addressing common limitations such as T-cell exhaustion. The addition of tislelizumab further blocks the PD-1/PD-L1 signaling pathway to sustain the anti-tumor immune response. This combination is primarily being developed by the Second Affiliated Hospital, School of Medicine, Zhejiang University, for the treatment of relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL).
