Overview

CD19 ALA-CART is an investigational chimeric antigen receptor (CAR) T-cell therapy developed by the University of Colorado Anschutz Medical Campus. It utilizes the proprietary Adjunctive LAT Activating CAR (ALA-CART) platform, which incorporates a bicistronic construct expressing both a standard second-generation CAR (typically with a 4-1BB costimulatory domain) and a novel adjunctive CAR containing the intracellular signaling domain of Linker for Activation of T cells (LAT). This design is intended to overcome signaling deficits in standard CARs, particularly in response to low antigen density, by enhancing LAT scaffolding and downstream signaling. Preclinical studies demonstrate that CD19 ALA-CART can eradicate CD19-low leukemia cells that are resistant to standard CD19-BBz CAR T cells, while maintaining a less differentiated stem cell memory phenotype and reducing exhaustion.
