CD19 allogeneic CAR-T is an investigational donor-derived chimeric antigen receptor (CAR) T-cell therapy developed by Tianjin First Central Hospital. The therapy involves engineering healthy donor T cells to express a CAR targeting the CD19 antigen, which is highly expressed on the surface of malignant B cells. The CAR construct includes a humanized anti-CD19 single-chain variable fragment (scFv) linked to 4-1BB and CD3ζ signaling domains to promote T-cell activation and persistence. As an allogeneic product, it is designed as an "off-the-shelf" treatment, potentially overcoming the manufacturing delays and cell quality issues associated with autologous CAR-T therapies. It is currently being evaluated in Phase 1/2 clinical trials for patients with relapsed or refractory B-cell malignancies, including acute lymphoblastic leukemia and non-Hodgkin lymphoma.
