Overview

CD19-ARM is a novel bispecific antibody known as an Activating Receptor Multifunctional (ARM) bispecific, developed by Janux Therapeutics[1]. It is designed as a T cell engager that targets CD19, a pan-B cell surface marker expressed on normal and malignant B cells[1][4]. The drug is engineered to bind both CD19 on B cells and CD3 on T cells, bringing them into close proximity to activate T cell-mediated killing of CD19-positive cells[1]. CD19-ARM is specifically designed for the treatment of autoimmune diseases by depleting B cells and resetting the immune system[1]. In preclinical studies, the therapy demonstrated complete B-cell depletion in peripheral blood and tissues with prolonged memory B-cell reset, notably without causing cytokine release syndrome (CRS)[1]. The drug is currently in IND-enabling studies and is intended to be a fully off-the-shelf, re-dosable therapeutic that can drive prolonged drug-free remissions[1].
