CD19-BAFF CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy that dual-targets the CD19 antigen and the B-cell activating factor receptor (BAFF-R, also known as TNFRSF13C). This bispecific approach is designed to address the limitations of single-target CAR-T therapies, particularly antigen escape, which is a frequent cause of relapse in patients treated with CD19-directed therapies. BAFF-R is a critical survival receptor expressed on the surface of most B-cell malignancies and mature B-cells, making it an ideal co-target. The therapy is currently being evaluated in clinical trials for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL) and B-cell non-Hodgkin lymphoma (B-NHL), as well as a range of B-cell-mediated autoimmune diseases including systemic lupus erythematosus (SLE), systemic sclerosis, and neuromyelitis optica. By simultaneously targeting two essential B-cell surface proteins, CD19-BAFF CAR-T cells aim to achieve more complete and sustained B-cell depletion.
