**CD19-BAFF-R dual CAR T cells** are a target-defined, bispecific CAR-T-cell therapy platform developed at City of Hope for B-cell malignancies, principally acute lymphoblastic leukemia. The engineered T cells co-recognize CD19 and B-cell activating factor receptor, using a dual-scFv CAR architecture with 4-1BB costimulation and CD3ζ signaling to kill malignant B cells expressing either antigen. In published preclinical work, the optimized loop-format construct eradicated mixed CD19-negative and BAFF-R-negative acute lymphoblastic leukemia xenografts, addressing antigen-escape relapse observed after monospecific CD19-directed CAR-T therapy.
