CD19-BAFF targeted CAR T-cells

CD19-BAFF targeted CAR T-cells are a dual-targeting chimeric antigen receptor T cell therapy designed to simultaneously target both CD19 and BAFF receptor (BAFF-R) on B-cell malignancies. This bispecific CAR construct was developed to mitigate antigen escape, which is a major cause of treatment failure in CD19-targeted CAR T therapies. The dual-targeting approach allows the therapy to remain effective even if one tumor antigen becomes downregulated during treatment, as the second targeting domain continues to be reactive to tumors. This is particularly valuable for treating patients with CD19-negative relapse after prior CD19-directed therapies, as BAFF-R expression is often retained in these cases.
