Overview

CD19-BCMA CAR-T cells are a type of autologous chimeric antigen receptor (CAR) T cell therapy engineered to simultaneously target both B-cell maturation antigen (BCMA) and cluster of differentiation 19 (CD19). These dual-targeted CAR-T therapies are designed to recognize and bind to tumor or pathogenic immune system cells expressing either or both antigens. Upon administration, the modified T-cells induce cytotoxicity against these target-expressing B-cells and plasma/long-lived plasma cells. This approach aims for a more comprehensive depletion of disease-causing B-lineage populations compared to single-targeted therapies. The primary indications under investigation include refractory systemic lupus erythematosus (SLE), lupus nephritis, and certain hematologic malignancies[2][3][4]. Early clinical studies suggest that this therapy can induce rapid and durable remission in severe autoimmune diseases by resetting the humoral immune system[3][4]. The mechanism involves direct cytolysis of BCMA+ plasma/plasma-like B-cells as well as CD19+ mature/immature B-cells.
