Overview

CD19-BCMA dual nanobody-based CAR-T cells is an experimental cellular immunotherapy designed for desensitization in patients with donor-specific antibodies (DSA) who are candidates for allogeneic hematopoietic stem cell transplantation. The therapy utilizes T cells engineered to express chimeric antigen receptors (CARs) that simultaneously target CD19, found on B cells, and B-cell maturation antigen (BCMA), found on plasma cells. By depleting both B cells and antibody-secreting plasma cells, the treatment aims to reduce or eliminate DSAs that would otherwise cause graft rejection or poor engraftment. This specific construct utilizes nanobody-based binding domains, which are smaller and potentially more stable than traditional single-chain variable fragments (scFvs).
