CD19-CAR-Lenti-ALLO is an allogeneic, donor-derived chimeric antigen receptor (CAR) T cell therapy targeting the CD19 antigen on B cells. It is manufactured by transducing donor T cells with a lentiviral vector encoding a second-generation CAR construct that includes an anti-CD19 single-chain variable fragment (scFv), a 4-1BB costimulatory domain, and a CD3ζ signaling domain. This therapy is designed for patients with relapsed or refractory B-cell precursor acute lymphoblastic leukemia (BCP-ALL), particularly those who have relapsed after allogeneic hematopoietic stem cell transplantation (allo-HSCT) or are ineligible for autologous CAR-T products due to profound lymphopenia or rapidly progressing disease. In clinical studies, it has demonstrated efficacy in inducing complete remission with minimal residual disease negativity and has shown a manageable safety profile comparable to autologous CAR-T therapies[1].
