Overview

CD19 CAR NK cells are natural killer (NK) cells that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD19 antigen, which is commonly found on the surface of B-cell malignancies such as non-Hodgkin lymphoma and chronic lymphocytic leukemia. The mechanism of action involves redirecting the cytotoxic activity of NK cells specifically toward CD19-expressing tumor cells. Unlike traditional T cell-based therapies, these allogeneic "off-the-shelf" cell therapies do not require HLA matching and are associated with minimal side effects such as cytokine release syndrome or neurotoxicity. Clinical studies have shown promising results in patients with relapsed or refractory B-cell malignancies, including high rates of complete remission and favorable safety profiles[1][2][3]. Some approaches combine memory-like properties induced by cytokines (IL-12/15/18) to enhance persistence and antitumor efficacy[3].
