Drug intelligence / Profile preview
CD19 CAR-T cells (third-generation) are autologous or allogeneic T cells genetically engineered to express a chimeric antigen receptor (CAR) targeting the B-cell antigen CD19. Unlike second-generation CARs, which incorporate a single costimulatory domain (typically CD28 or 4-1BB), third-generation CARs feature two costimulatory domains (such as CD28 and 4-1BB, or CD28 and TLR2) in tandem with the CD3-zeta signaling domain. This dual costimulatory design is intended to enhance T-cell activation, proliferation, and in vivo persistence, potentially improving therapeutic efficacy and durability in patients with B-cell malignancies, including leukemia and lymphoma, while maintaining a manageable safety profile.
