Overview

**CD19 CAR-T with membrane-bound interleukin-15** is a target-specific armored CAR-T-cell platform represented clinically by the autologous construct CAR19-41BB-CD3ζ-mIL15-T cells. The engineered T cells express an anti-CD19 CAR incorporating a CD8 hinge and transmembrane region, 4-1BB costimulatory domain, and CD3ζ signaling domain, together with membrane-bound IL-15 linked through a P2A sequence. CD19 recognition directs cytotoxic T-cell activity against CD19-positive B-lineage malignant cells, while membrane-bound IL-15 is intended to promote CAR-T expansion, survival, memory-like persistence, and antitumor function. A published case report from The Fifth Medical Center of Chinese People's Liberation Army General Hospital described complete remission with incomplete hematologic recovery and minimal residual disease negativity for approximately five months in a patient with multiply relapsed B-cell acute lymphoblastic leukemia after infusion of this construct.
