Overview

CD19-CAR_Lenti T cell is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy developed by the Bambino Gesù Hospital and Research Institute. The therapy consists of T cells genetically modified via a self-inactivating lentiviral vector to express a CAR targeting the CD19 antigen, which is found on the surface of B cells. The construct includes a 4-1BB costimulatory domain to enhance the persistence and anti-tumor efficacy of the infused cells. Manufactured using the automated Miltenyi CliniMACS Prodigy system, the therapy is designed for point-of-care production with a rapid vein-to-vein time of approximately 14 days. It is currently being investigated in Phase I/II clinical trials for the treatment of pediatric and young adult patients with relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL), diffuse large B-cell lymphoma (DLBCL), and primary mediastinal B-cell lymphoma (PML).
