CD19-CD22-bispecific CAR T cells is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed by St. Jude Children's Research Hospital for the treatment of pediatric patients with relapsed or refractory B-cell acute lymphoblastic leukemia (ALL). The therapy involves genetically engineering a patient's own T cells to express a bispecific CAR that simultaneously targets two B-cell surface antigens: CD19 and CD22. This dual-targeting approach is designed to mitigate the risk of antigen escape, a common cause of relapse in single-target CAR T therapies where leukemia cells downregulate or lose the target antigen. The treatment process includes lymphodepleting chemotherapy followed by a single infusion of the modified T cells. It is currently being evaluated in a Phase 1 clinical trial (1922CAR) for safety and efficacy in patients aged 21 and younger.
