CD19-CD34 CAR transduced T cells are autologous (patient-derived) T lymphocytes that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting the B-cell surface antigen CD19, with an added CD34 selection marker. The CAR construct enables these modified T cells to recognize and kill malignant B cells expressing CD19, a protein commonly found on the surface of most B-cell malignancies such as acute lymphoblastic leukemia (ALL) and certain types of lymphoma. The inclusion of the CD34 marker allows for enrichment or selection of successfully modified T cells during manufacturing. This therapy is being investigated as an immunotherapy for patients with recurrent or refractory B cell malignancies who have failed standard treatments. The mechanism involves direct cytotoxicity against cancerous B-cells via recognition and binding to the CD19 antigen, leading to tumor cell lysis[1][2][3][4].
