CD19-CD34t metabolically programmed CAR transduced T-cells is a novel cellular immunotherapy currently being investigated in clinical trials. This therapy represents an innovative approach in the field of CAR T-cell therapy, specifically designed to address some of the limitations of current CD19 CAR T-cell treatments. This therapy incorporates a CD34 tag (CD34t) into the CAR T-cell construct, which allows for a more purified CAR T-cell product through CD34 selection. The purification step is intended to improve safety and reduce toxicity compared to conventional CAR T-cell therapies. The cells are metabolically programmed to enhance their fitness, addressing one of the key issues with current CAR T-cell therapies - relapse due to lack of CAR T-cell fitness. The collected T-cells are exposed to priming conditions that lead to a metabolically enhanced CAR-T cell product similar to a Th1/17 hybrid cell. This approach aims to yield a high-quality product with enhanced persistence and anti-tumor efficacy when purified based on CD34t expression.
