Overview

CD19 chimeric antigen receptor (CAR) T cell therapy is a form of cellular immunotherapy where a patient's T cells are genetically engineered to express a synthetic receptor that recognizes the CD19 antigen, which is widely expressed on B-cell lineage cells. Upon infusion, these modified T cells target and eliminate CD19-positive malignant cells. This modality has revolutionized the treatment of relapsed or refractory B-cell malignancies, including acute lymphoblastic leukemia (ALL), diffuse large B-cell lymphoma (DLBCL), and mantle cell lymphoma (MCL). Common toxicities include cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS), as well as increased susceptibility to bacterial, viral, and fungal infections due to lymphodepletion and B-cell aplasia.
