A specialized cellular therapy product developed by St. Jude Children's Research Hospital for use in allogeneic hematopoietic cell transplantation (HCT). The product consists of donor-derived hematopoietic progenitor cells that have undergone ex vivo depletion of CD19+ B-lymphocytes, typically using the CliniMACS magnetic cell separation system. This depletion is performed to minimize the risk of post-transplant lymphoproliferative disorder (PTLD) caused by Epstein-Barr virus (EBV) reactivation in the donor B-cells. In the context of haploidentical HCT for pediatric patients with high-risk hematologic malignancies (such as ALL and AML), this CD19-depleted graft is often administered alongside TCRαβ-depleted cells to reduce the incidence of graft-versus-host disease (GVHD) while maintaining a potent graft-versus-leukemia effect through preserved NK cells and γδ T-cells.
