Overview

CD19-directed CAR-T cell therapy is a form of adoptive cell immunotherapy where a patient's T cells are genetically modified to express a chimeric antigen receptor (CAR) specific for the CD19 antigen, a protein expressed on the surface of most B-cell malignancies. Upon re-infusion, these engineered cells recognize and eliminate CD19-positive tumor cells. In clinical research conducted by The Second Xiangya Hospital of Central South University, advanced iterations of this technology are being evaluated, including CAR-T cells engineered to express IL-18 to enhance anti-tumor activity and persistence, as well as versions where IL-6, IL-1B, and GM-CSF are silenced to potentially mitigate cytokine release syndrome (CRS) and other associated toxicities.
