CD19-directed CAR-T cells developed by the Chinese PLA General Hospital are an investigational allogeneic (donor-derived) chimeric antigen receptor T-cell therapy. This specific treatment is being evaluated in Phase I clinical trials for patients with relapsed or refractory B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL) and B-cell lymphoma. The therapy involves engineering T cells from a healthy donor to express a CAR targeting the CD19 antigen, which is widely expressed on B-cell lineage cells. In the clinical protocol described, these CAR-T cells are administered prior to a donor-derived hematopoietic stem cell transplant (HSCT) to eradicate residual disease and potentially enhance the graft-versus-leukemia (GVL) effect while minimizing the risk of CD19-negative relapse.
