CD19-FLAG CAR-T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the CD19 protein, which is highly expressed in B-cell malignancies. These cells are genetically modified to express a CAR that includes a FLAG octapeptide tag (DYKDDDDK) within its extracellular domain. This tag serves as a molecular marker, allowing for the precise detection, enrichment, and monitoring of the CAR-expressing T cells using anti-FLAG antibodies without compromising the CAR's affinity for CD19 or its therapeutic potency. In preclinical studies, CD19-FLAG CAR-T cells have demonstrated significant efficacy in eliminating CD19-positive hematological cancer cells, such as Raji Burkitt's lymphoma, as well as solid tumor models engineered to express CD19 (e.g., HeLa-CD19). The therapy works by inducing T-cell activation, proliferation, and the secretion of pro-inflammatory cytokines like IFN-gamma and IL-2 upon antigen recognition, leading to targeted tumor cell lysis and extended survival in animal models.
