Overview

CD19 HIT T-cells are an investigational **autologous engineered T-cell therapy** in which patient T cells are modified to express a **human leukocyte antigen-independent T-cell receptor** directed against **CD19**. HIT receptors are synthetic receptors that reconfigure antigen recognition through the endogenous **T-cell receptor CD3 signaling complex** rather than a conventional CAR architecture, with the goal of increasing antigen sensitivity and enabling recognition of tumor cells with lower target density. Preclinical work from Memorial Sloan Kettering investigators, including the Sadelain laboratory, showed that CD19-directed HIT T cells can mediate potent anti-leukemia activity and may outperform matched CD28-based CAR designs in low-antigen settings. The program appears to be a **research-stage cell therapy construct** rather than an approved or broadly branded clinical product, and publicly available information is primarily preclinical.
